Pseudohypoparathyroidism in childhoodArticles

Pseudohypoparathyroidism (PHP) is characterised by biochemical hypoparathyroidism with elevated parathyroid hormone (PTH) levels and reduced target tissue responsiveness to PTH. Patients with PHP type 1a show a group of developmental defects termed Albright hereditary osteodystrophy (AHO) and exhibit resistance to additional hormones because of heterozygous, inactivating mutation in GNAS1, the gene for the alpha-subunit of the Gs-protein, which disrupts Gs-protein-coupled signal transduction pathways. Within PHP type 1a families some individuals show AHO but have normal hormone responsiveness, a variant phenotype termed pseudo-pseudohypoparathyroidism...

Strontium renelate - a prospective compound for the treatment of osteoporosisArticles

In vitro studies gave evidence that strontium ranelate stimulates bone formation and has an inhibitory effect on bone resorption. Preclinical studies on animal models of osteoporosis further confirmed that application of strontium leads to an increase in bone mass and has a preventive effect on bone loss. Placebo-controlled phase II studies proved that 1 to 2 years treatment with strontium ranelate in daily doses of 1 000-2 000 mg resulted in significant increase in bone mineral density. Phase III studies gave evidence that treatment with daily doses of 2 000 mg strontium ranelate signifi­ cantly reduced both vertebral and nonvertebral fracture risk....
